Chauffered Chopper heirloom mount coming in patch 6.1
Enter the Chauffered Chopper, which is currently on the patch 6.1 test realm. Like the name implies, this is a motorcycle-style mount with a driver (currently orc Koak Hoburn, likely a nod to Driving Miss Daisy chauffeur Hoke Colburn). Having your own chauffeur means that you can ride around in this chopper at any level, making it a must-have for alt-a-holics on the move. You can pick up your own by nabbing the Heirloom Hoarder achievement for collecting 35 heirlooms.
The mount isn't currently working on the PTR, so it appears to be a work in progress. Also, it's more than a little odd to have an orc chauffeur for a distinctly alliance-styled mount, so we wonder if its look might be changing before it goes live. We'll keep an eye on the PTR for more info when it's there!

Manchester City Maintain Title Hopes at Chelsea, but Draw Is 2 Points Dropped
Manchester City became the first side to take anything away from Stamford Bridge in the Premier League this season after a 1-1 draw with leaders Chelsea, but they will leave the capital with a feeling of what could have been.
It’s a result that leaves Jose Mourinho’s side five points clear at the top of the table but ends their run of 10 consecutive home wins, which has provided the platform for their strong first half of the season.
And they will be relieved to escape with their five-point lead in tact, after City dominated for long periods. Without Cesc Fabregas, who has 15 assists in the league already this season, and Diego Costa, their 17-goal striker, Chelsea looked shaky and bereft of quality.
They only managed two attempts on goal—none after the 41st minute—as City looked by far the more likely winners. Manuel Pellegrini’s side, who have still only lost once on the road all season, will see this as two points dropped, but there’s still plenty to play for in this title race.
It was a much improved performance from City, who have struggled to produce their best since the boxing Day win over West Bromwich Albion, and provides them with a significant boost for the coming weeks.
Their improvement was best exemplified by James Milner and Jesus Navas, both of whom were excellent throughout.
In the recent defeat to Arsenal, Milner was taken off at half time after an ineffective display, but here he was at his very best, harrying, tackling and forcing mistakes from opponents, offering attacking thrust and defensive solidity.
And Navas, who hit 18 failed crosses against the Gunners and has faced criticism of his big-game ability for some time, gave Cesar Azpilicueta his most difficult 90 minutes of the season, constantly looking to beat him, either by playing one-twos with his team-mates or using his pace to go beyond the Chelsea full-back.
Chelsea took the lead shortly before half time, after a first-time cross from Eden Hazard found Loic Remy inside the box and the Frenchman finished past Joe Hart, coming somewhat against the run of play.
Vincent Kompany, who was much improved from his display against Middlesbrough in the FA Cup, could probably have dealt with the cross, but wary of putting the ball in his own net, he decided to allow it to go beyond him. Remy couldn't miss.
It was a sickener for City, who had edged proceedings in the opening period, but they found an equaliser moments before the break.
A superb cross from Navas caused Thibaut Courtois problems, as he flapped at the ball and allowed it to fall at the feet of Sergio Aguero. The Argentinian smashed it left-footed towards goal, leaving David Silva to apply the crucial touch and bring the sides level.
City’s marginal domination grew more pronounced after the break, with Chelsea clearly struggling after their gruelling midweek League Cup semi-final with Liverpool had entered extra time.
They looked out on their feet, struggling to keep pace with their opponents, and City, sensing a seismic victory, were by far the dominant side, but they couldn’t quite find what would have been a hugely significant winning goal.
There are positives for City, though. Traditionally, they find their best form towards the end of the season, and with Yaya Toure, Samir Nasri and Wilfried Bony all set to come in and strengthen their side, they will only get better after what’s been a difficult few weeks.
After allowing their standards to slip, this game signalled a return to form for the Citizens, and the belief it will give them could be a launchpad for another attempt at eroding Chelsea's lead.
However, this could so easily have been a momentum-shifting win. Pellegrini surely sees this as a lost opportunity.

Patch 6.1: Traveling the world in search of garrison music
The jukebox is one of the biggest new garrison features coming in patch 6.1. It allows you to change the default music in your garrison, which is a welcome feature considering how much time I spend in there. Upon completing the simple quest to build your jukebox, you are given a selection of 7 faction-specific music tracks (known as Music Rolls) to get you started. It's as simple as clicking your jukebox and picking a new track as your default garrison music.
There are many more Music Rolls to collect. One of the first I collected on the PTR was Music Roll: Faerie Dragon, which I found all the way out in Tirisfal Glades. If you're a regular reader of Know Your Lore, you may remember the Tinfoil Hat Edition about the mysterious Whispering Forest event (pictured above). If you are present for this event -- which has been made less rare in patch 6.1 -- you can loot this music roll from a faerie dragon nest located in the center of the circle of mushrooms.

Vitamin D levels and status amongst asthmatic and non-asthmatic adolescents in Cyprus: a comparative cross-sectional study
Emerging evidence suggests that vitamin D might be implicated in asthma pathophysiology. This study aims to compare Vitamin D mean serum levels and status between asthmatic and non-asthmatic adolescents and investigate the association of vitamin D with asthma severity.
Methods:
In a cohort of adolescents aged 16?17 years, those reporting wheezing in the past 12?months and Ever asthma on the ISAAC questionnaire were invited to participate and formed the Active Asthmatics group. Controls were selected amongst Never Wheezers/Never Asthmatics (NWNA). Differences in mean 25(OH)D serum levels and vitamin D status between AA and NWNA were examined in multivariate linear and logistic regression models respectively, adjusting for potential confounders. Within AA, differences in vitamin D levels were assessed across asthma severity indicators.
Results:
A total of 69 AA and 671 NWNA participated in the study. Unadjusted mean 25(OH)D serum levels were 22.90 (SD 6.41), and 21.15 (SD 5.59) ng/mL in NWNA and AA respectively (p?=?0.03). In adjusted models, mean 25(OH)D levels remained significantly lower amongst AA compared to NWNA (adjusted beta coefficient ?1.68, 95%CI ?3.24, ?0.13). Severe (<12?ng/mL), moderate (<25?ng/mL) or insufficient (<30?ng/mL) vitamin D status was more prevalent among AA who were 1.6 times (95% CI 1.01, 2.53) more likely to belong to a lower vitamin D category compared to NWNA. Within AA, there was a negative trend between vitamin D levels and the number of reported asthma severity indicators. Conclusions: Levels of vitamin D tend to be lower among asthmatic compared to non-asthmatic children and in those with severe asthma independent of important confounders.

Chris Weidman vs. Vitor Belfort Out, Rousey vs. Zingano in at UFC 184 Main Event
Chris Weidman will not defend his title against Vitor Belfort on Feb. 28 at UFC 184.
According to MMAJunkie.com, the UFC middleweight champion, Weidman, has suffered an injury in training that forced him off the card.
In the wake of this news, a women's bantamweight title fight between Ronda Rousey and Cat Zingano, which was originally scheduled as the show's co-main event, will receive top billing on the evening.
Per MMA Junkie's report, neither the nature of Weidman's injury nor a timetable for his return is known at this time.
For Weidman, unfortunately, injuries are nothing new.
In September 2014, he was forced to pull out of a UFC 181 title showdown against Belfort due to a broken hand. He's also previously injured his shoulder and his knee while fighting under the UFC banner.
While Rousey vs. Zingano is an awesome fight that will provide the women's bantamweight champ with perhaps her toughest test to date, this is a massive blow to the Los Angeles card.
Weidman vs. Belfort was a killer main event, and a one-two punch of excellent matchups with title implications looked great for the promotion.
Now, the co-main event for the evening will become Josh Koscheck vs. Jake Ellenberger, a showdown between aging welterweights with absolutely no bearing on the title picture at 170 pounds.
In fact, both men are coming off three straight losses inside the Octagon and are perhaps fighting for their careers. While it's a fun novelty match between well-known names, it has absolutely nothing on Rousey vs. Zingano as a co-main event in the grand scheme of things.

Titans Announce Changes to Front Office: Latest Details, Comments and Reaction
With the 2015 NFL draft still a few months away, the Tennessee Titans decided the time is now for a major front-office shake-up.
The team announced on Friday that it parted ways with Lake Dawson, Tennessee's now former vice president of player personnel, in addition to a few tweaks to the management structure. The Titans posted a statement from general manager Ruston Webster on Twitter:
The decision to let Dawson go caught quite a few NFL fans and insiders off guard. Albert Breer of NFL Network noted that he was a general manager candidate for a few different teams in the past few years:
Dan Phillips of Fox 17 in Nashville hinted that the team is scapegoating Dawson to cover for larger issues in the organization:
Dawson had spent the last eight seasons inside the Titans organization, starting out as the director of pro personnel in 2007 before assuming his most recent role in 2011. He worked in the Seattle Seahawks' personnel department before arriving in Nashville.
The Titans haven't made the playoffs since 2008 and have just one winning season during that time. After last year's 2-14 record, many fans in Tennessee will agree that something needs to change in the front office for the franchise to be competitive again.
Ultimately, though, Tennessee will need to improve its on-field product, and that starts with the draft. The Titans have the second overall pick in the first round, and it's imperative that they nail the selection.
Letting Dawson go is likely a part of the franchise's long-term plans.

Time-to-infection by Plasmodium falciparum is largely determined by random factors
The identification of protective immune responses to P. falciparum infection is an important goal for the development of a vaccine for malaria. This requires the identification of susceptible and resistant individuals, so that their immune responses may be studied. Time-to-infection studies are one method for identifying putative susceptible individuals (infected early) versus resistant individuals (infected late). However, the timing of infection is dependent on random factors, such as whether the subject was bitten by an infected mosquito, as well as individual factors, such as their level of immunity. It is important to understand how much of the observed variation in infection is simply due to chance.
Methods:
We analyse previously published data from a treatment-time-to-infection study of 201 individuals aged 0.5 to 78�years living in Western Kenya. We use a mathematical modelling approach to investigate the role of immunity versus random factors in determining time-to-infection in this cohort. We extend this analysis using a modelling approach to understand what factors might increase or decrease the utility of these studies for identifying susceptible and resistant individuals.
Results:
We find that, under most circumstances, the observed distribution of time-to-infection is consistent with this simply being a random process. We find that age, method for detection of infection (PCR versus microscopy), and underlying force of infection are all factors in determining whether time-to-infection is a useful correlate of immunity.
Conclusions:
Many epidemiological studies of P. falciparum infection assume that the observed variation in infection outcomes, such as time-to-infection or presence or absence of infection, is determined by host resistance or susceptibility. However, under most circumstances, this distribution appears largely due to the random timing of infection, particularly in children. More direct measurements, such as parasite growth rate, may be more useful than time-to-infection in segregating patients based on their level of immunity.

Why the Chicago Bears Absolutely Must Retain Lance Briggs
The Chicago Bears have 20 unrestricted free agents entering the 2015 offseason, and none are more important to retain than Lance Briggs.
What looked like a closed door has now cracked opened with a new regime in town. Briggs recently spoke with Brad Biggs of the Chicago Tribune, expressing his willingness to return.
"New GM, new coach, we haven't had those kinds of talks yet. Obviously the door is open until we have that conversation and I know how they feel," said Briggs.
By now, most tend to think Briggs has worn out his welcome in Chicago and has one foot out the door, but he still has some good football left in him and should spend the final years of his career with the Bears.
After all, Briggs could end up being the next Julius Peppers.
Peppers had a down season in his final year as a Bear. He seemed to dog it on the field and looked like age had caught up to him. Fast forward a year and Peppers looks fresher than ever in a Packers uniform.
What caused Peppers' turnaround? He latched onto a quality organization with a respectable coach.
Doesn't all of this sound familiar with Briggs? It's clear former head coach Marc Trestman didn't get the respect he needed from players, and the veterans on defense seemed to regress under his watch.
If Briggs were to leave Chicago for let's say Tampa Bay, he would likely look focused and rejuvenated under a respected coach like Lovie Smith.
Head coach John Fox and defensive coordinator Vic Fangio can get quality play out of Briggs again. Don't pay attention to scheme, Briggs can play in a 3-4 if needed. Former Bears All-Pro linebacker Brian Urlacher seems to think so as well.
"I think Lance [Briggs] fits great in any defense," said Urlacher. "His instincts make him a great fit for any defense. The way he reads things, he reads things like a running back, which is good in a 3-4."
In a 3-4 scheme, the defensive linemen and linebackers play more two-gap technique. This requires more discipline as they are responsible for the A and B gaps.
Briggs' greatest strength in his career has been his ability to read and react to plays quickly. The Bears can line him up inside next to Jon Bostic to help the young defender read the gaps.
An underrated skill in Briggs' career is his ability to blitz. He has 15 sacks in his career and could be a menace once again from the middle or outside.
Over the past two years, Briggs has played in only 17 games due to various injuries. As long as he's healthy, it's a low-risk, high-reward move to bring Briggs back on a one-year deal.
If the Bears re-sign Briggs, it doesn't cost much money and they can still bring in a young impact player without having to throw him into the fire right away.
The organization already fumbled when they mishandled Urlacher's situation, and Briggs likely wants to put his focus back on football, since his Double Nickel Smokehouse in California is currently averaging 2.5 stars on Yelp.
General manager Ryan Pace is going to set a foundation based on success, hard work and pride.
Regardless of what has happened over the last few years, Briggs still embodies all of those qualities. Bring him back for a year, let him finish his career in Chicago and allow him to be a meaningful bridge to a new generation of good Bears linebackers.

Projecting the Cincinnati Reds' 2015 Opening Day Roster
At this point in the MLB offseason, most teams have made all of their impact signings and trades. That means rosters are beginning to take shape.
The Cincinnati Reds made their fair share of moves this winter. Gone are Mat Latos, Chris Heisey and Alfredo Simon; Marlon Byrd was the team's biggest acquisition of the offseason.
With those moves, Cincinnati's roster is going to have a different look to it in 2015. Although there will be a few roster spots up for grabs in spring training, the team's core is good enough to contend for a playoff spot if it stays healthy.

The first of two major ACC showdowns featuring the No. 4 Duke Blue Devils tips off at 7:30 p.m. ET at Purcell Pavilion.
The Blue Devils, who are coming off an emotional Sunday win over St. John's, will visit the No. 8 Notre Dame Fighting Irish three days before their trip to Virginia.
Although they haven't received a ton of national attention, the Fighting Irish are 7-1 in conference play with their loss coming at the hands of Virginia.
The big name to watch Wednesday night is Notre Dame's Jerian Grant, who was shut down in the loss at home to Virginia. Outside of that game and the win over North Carolina, Grant has scored in double digits in every game for Mike Brey's team.
On the other side of the court, all eyes will be on Jahlil Okafor, who is averaging 18.5 points and 9.0 rebounds per game in his freshman season.

WWE Royal Rumble 2015: Contests That Lived Up to the Hype
WWE's Royal Rumble event is in the books, and fans are likely still buzzing about the final match of the night. That match saw Roman Reigns emerge victorious as the final Superstar in the Rumble, guaranteeing him a WWE World Heavyweight Championship shot at WrestleMania 31.
And while many are likely still debating the pros and cons of that move, the fact is that the rest of the Rumble card had some definite bright spots. Indeed, there were several moments that definitely lived up to the hype.
The Ascension vs. The New Age Outlaws
Many fans were likely not expecting too much from this match. The reason for that involves WWE's newest duo, The Ascension, which came out of hiding in Florida to totally underwhelm on the company's main roster.
The truth is, Konnor and Viktor were put squarely behind the eight ball when they began on Monday Night Raw.
Fans that knew them from NXT were surely happy to see them finally make it to WWE's flagship program. Those that didn't know them perhaps saw just another tag team on the scene, one that would probably be mixing it up with The Usos at some point.
But rather than play them as a powerhouse tag team with championship ambitions, WWE instead chose to portray The Ascension as rip-offs of The Road Warriors.
It was their primary talking point, one they used from the beginning. Instead of letting the crowd decide for themselves what to make of The Ascension, the comparisons to Hawk and Animal were forced onto the WWE faithful every time the new team was on the air.
So seeing The Ascension at the Royal Rumble was likely a moment that many were anticipating, if only to see how the pair would respond under the bright lights. But the fact is they looked surprisingly good. Of course, they were in the ring with the veteran New Age Outlaws, and it would actually be a little difficult to have a bad match under those circumstances.
But The Ascension gave as good as they got and finally looked like a hardworking tag team instead of a forced mimicked gimmick. This was the duo's chance to shine, and they took full advantage of it.
The Royal Rumble match
This may be a tough one for many fans, especially considering that the aforementioned Reigns walked out with the win and the hatred of the crowd. But there were other workers in the match that had some nice moments—moments that were far less controversial than the final one.
Diamond Dallas Page surprisingly entered the Rumble, the first of his pro wrestling career. DDP came in and looked as though he fit right in with the roster, carrying his weight against several Superstars. He did his signature diamond salute to the crowd, hit the Diamond Cutter and popped the crowd quite a few times during his run in the match. It was a nostalgic appearance but one that fans responded very positively to.
Bubba Ray Dudley also returned to the WWE ring during the Rumble match. The former WWE Superstar also fit right in again among his peers and even executed double-team moves with R-Truth that are usually reserved for Devon Dudley. The Rumble emanated from ECW country in Philadelphia, so Bubba got a huge reaction, and that equated to him delivering on every level in the ring.
Dean Ambrose was a favorite of many to win the Rumble and move on to WrestleMania 31, but of course, that did not happen. That does not, however, negate from the impact he made in the match and how good he looked against the field. He showed once again that he belongs on the main event stage, and this will surely not be the last time he appears on it.
Brock Lesnar vs. John Cena vs. Seth Rollins
Reigns going over in the Rumble may have been a foregone conclusion to many fans, but the winner of the WWE World Heavyweight Championship match was not. There were many variables in this one, and they all began with Seth Rollins.
Rollins was perhaps the underdog to win the belt. After all, it was the first time he stepped into the ring with Brock Lesnar. And considering John Cena's status as the company's top guy, there was every reason to believe that Rollins did not stand a chance.
But he stepped up in the match. Rollins held his own and looked good against The Beast Incarnate. Despite the size difference and the difference in the main event-level experience, Rollins proved he deserved the spot he had. It was the biggest match of his life, and Rollins delivered in a very big way.
Cena and Lesnar also came through on their part of the bout, as both men brought the intensity that they've been known for in the past. For the first time since he won the belt, Lesnar looked like the champion that WWE needed. Gone was the doubt and hatred from fans. The only thing that mattered was the three men in the ring and Paul Heyman living the emotion of it all on the outside.
This was the kind of championship match that WWE fans had been waiting on, the kind that could get them on their feet and remind them of how entertaining a big main event could be. It also set the wheels into motion for the future of the company.
The title is only as good as the focus that it receives from WWE. Until the strap comes back to a full-time performer, fans need to see the company give it proper respect, and the only way to do that is to feature it in high-profile matches with top-notch stars. And that is exactly what happened at the Royal Rumble.

Bayesian models for syndrome- and gene-specific probabilities of novel variant pathogenicity
With the advent of affordable and comprehensive sequencing technologies, access to molecular genetics for clinical diagnostics and research applications is increasing. However, variant interpretation remains challenging, and tools that close the gap between data generation and data interpretation are urgently required. Here we present a transferable approach to help address the limitations in variant annotation.
Methods:
We develop a network of Bayesian logistic regression models that integrate multiple lines of evidence to evaluate the probability that a rare variant is the cause of an individual?s disease. We present models for genes causing inherited cardiac conditions, though the framework is transferable to other genes and syndromes.
Results:
Our models report a probability of pathogenicity, rather than a categorisation into pathogenic or benign, which captures the inherent uncertainty of the prediction. We find that gene- and syndrome-specific models outperform genome-wide approaches, and that the integration of multiple lines of evidence performs better than individual predictors. The models are adaptable to incorporate new lines of evidence, and results can be combined with familial segregation data in a transparent and quantitative manner to further enhance predictions.Though the probability scale is continuous, and innately interpretable, performance summaries based on thresholds are useful for comparisons. Using a threshold probability of pathogenicity of 0.9, we obtain a positive predictive value of 0.999 and sensitivity of 0.76 for the classification of variants known to cause long QT syndrome over the three most important genes, which represents sufficient accuracy to inform clinical decision-making. A web tool APPRAISE [http://www.cardiodb.org/APPRAISE] provides access to these models and predictions.
Conclusions:
Our Bayesian framework provides a transparent, flexible and robust framework for the analysis and interpretation of rare genetic variants. Models tailored to specific genes outperform genome-wide approaches, and can be sufficiently accurate to inform clinical decision-making.

Brock Lesnar vs. John Cena vs. Seth Rollins: WWE Title Match Saved Royal Rumble
Monday’s edition of Raw emanated from WWE headquarters in Stamford, Connecticut, due to the winter storm that smashed into the east coast. Instead of cancelling the show, the company decided to air the entire Triple Threat match from the 2015 Royal Rumble for the WWE Championship as part of the broadcast.
Most of the fans who witnessed the Royal Rumble pay-per-view live understood how great the Triple Threat was at the time, but after watching the match again, it was clear that the bout had saved the PPV from being regarded as one of the most disappointing in recent history.
The success of the WWE Championship bout was due largely to Brock Lesnar. The former NCAA collegiate wrestling and UFC heavyweight champion is an absolute physical freak, and his ability in the ring is unparalleled. After watching him throw challengers John Cena and Seth Rollins around like rag dolls, there is no questioning the raw strength of The Beast.
One aspect of Lesnar's game that doesn't get nearly as much credit is his ability to sell. Whether it was making Cena or Rollins look strong, Lesnar has the ability to help the WWE Universe believe his opponents have a shot to win against him by selling moves properly.
The man who truly stole the show Sunday night was Rollins. This was his first real opportunity as a singles competitor in the WWE to shine on the big stage. With two of the biggest names in the business around him, Rollins looked extremely confident, and it showed throughout the match.
Using huge moments that included diving from the top turnbuckle through the Spanish announce table and hitting the Phoenix Splash on Cena, Rollins made it clear that he was the most gifted technical wrestler in the match and one of the top high-flyers on the roster. Add in his ability to react to the contrasting styles of Lesnar and Cena on the fly, and Rollins proved the future of the WWE is very bright.
As much as some fans don't like him, another major reason this Triple Threat was successful is Cena. The 15-time champion has the ability to rise to any occasion and bring his performance level to the caliber of the challengers he is facing. With two of the best in Rollins and Lesnar standing in front of him, Cena understood the magnitude of the match and shined bright.
Cena’s versatility was on full display, and his willingness to make both look strong at times is one of the reasons he has been on top of the mountain for so long. There is no doubt he got plenty of offense in during the bout, but Cena made it clear that this match was a vehicle to further Lesnar as a dominant champion and Rollins as the future face of the company.
With each participant in the Triple Threat taking this opportunity and running with it—and the correct wrestler earning the victory in exciting fashion—this was an almost perfect match. The crowd in Philadelphia loved this bout, and the fans sitting at home were treated to a fully immersive Royal Rumble moment.
While the 30-man Battle Royal that closed the PPV disappointed many in the WWE Universe, the Triple Threat match for the WWE Championship will be regarded as one of the best of the year and the saving grace of this event.
